What the research actually shows
Colombo and colleagues observed 10 healthy controls and 22 patients with brittle nails. Among eight patients whose specimens aligned with treatment, 2.5 mg/day biotin was followed by a 25% objective increase in thickness; the remaining 14 had a nonsignificant 7% increase. Hochman and colleagues retrospectively evaluated 35 treated patients and reported subjective improvement in hardness or splitting in 22, or 63%. Chiavetta 2019 randomized 50 people to hydroxypropyl chitosan lacquer alone or lacquer plus biotin 10 mg/day; both groups improved and the combination showed a larger signal, but no biotin-only or untreated group separated the supplement's effect from the lacquer. Baseline deficiency was not established.

The 25% objective thickness increase came from eight uncontrolled paired cases, and subjective improvement in hardness or splitting came from 22 of 35 uncontrolled retrospective cases. The 2019 randomized study added biotin to an active nail lacquer and therefore could not isolate biotin.
