Calcium alpha-ketoglutarate,
does it really help with Reduction in biological age and lifespan extension?

30-Second Summary
?
Evidence Grade ? · Safety unknown
Combination-product observational signals exist, but human efficacy results for Ca-AKG alone are absent, so the judgment is deferred
What the
research shows
No human efficacy result is available to judge Ca-AKG alone for reducing biological age or extending lifespan. The 42-person report of an approximately eight-year reduction was an uncontrolled retrospective analysis of Rejuvant, which combines Ca-AKG with vitamins, while the pure Ca-AKG ABLE RCT has published only a protocol and recruitment report. General AKG use was not significant in the 4,260-person cross-sectional dataset, so the judgment is deferred.
What the
ads claim
Marketing links lower DNA-methylation-clock values to reversal of actual age and lifespan extension. Current human data concern a calculated biological-age surrogate, not disease incidence, functional independence, mortality, or lifespan.
*

Useful facts when choosing a product

  • The observational studies evaluated a delayed-release combination containing Ca-AKG and vitamins.
  • The retrospective before-and-after analysis included 42 people with about seven months of average use.
  • The planned ABLE RCT dose is delayed-release Ca-AKG 1 g/day.
  • No result directly measuring human lifespan extension was found.

01

What the research actually shows

Demidenko 2021 compared TruAge DNA-methylation age before and after four to ten months of Rejuvant, a combination of Ca-AKG and vitamins, in 42 people and reported an average reduction of about eight years, but the analysis was uncontrolled and retrospective. The Sandalova 2023 and Lim 2025 ABLE publications concern the protocol and recruitment of a pure Ca-AKG RCT, not efficacy results. In the Pabis 2026 cross-sectional cohort of 4,260 people, an association was observed for a delayed-release Ca-AKG combination, whereas general AKG use was not significant.

02

Why this is classified as ?

Positive observational evidence concerns a Ca-AKG-plus-vitamin combination, the pure Ca-AKG ABLE RCT has no efficacy result, and the association for general AKG was not significant; human efficacy for Ca-AKG alone is therefore ungraded.

Counterpoint. The association with DNA-methylation age remains a hypothesis for randomized follow-up. This judgment does not convert animal lifespan data into human lifespan evidence.

Rejudgment record. Reassessment (cross-check reflected) — Positive evidence is an uncontrolled retrospective analysis of a Ca-AKG-plus-vitamin combination; pure Ca-AKG has no ABLE efficacy result, and general AKG use was not significant cross-sectionally
